چکیده

It is well known that the physiological activities of compounds are affected by their molecular structures. Hence, it is very important for the risk assessment of environmental chemicals such as PCDD and PCDF to evaluate structure-activity relationships (SAR). For precise analysis of SAR, the compounds should be stereoselectively synthesized. In this paper, we evaluated SAR of flaxseed lignan, secoisolariciresinol (SECO). SECO has many physiological effects such as immunostimulatory activity, antioxidant activity, and estrogenic activity. In addition, we first achieved the stereoselective synthesis of meso-SECO and optically active SECOs. Hence, SECO is a very good example for examining SAR. Optically active (+)and (–)-SECO, and meso-SECO were synthesized and various physiological functions were compared. At first, the immunostimulatory effects were examined. (+)and (–)-SECO accelerated IgM production of human hybridoma cells, however, meso-SECO did not. However meso-SECO showed cytotoxic activity against human breast cancer MCF-7 cells, (–)-SECO slightly stimulated the cell. This means that estrogenic activity of SECO derived from only (–)-SECO. Although (–)-SECO stimulated adiponectin production of 3T3-L1 adipose cells, (+)and meso-SECO suppressed the production. These facts suggested that slight difference in the structure of SECO affects the physiological activity.

The influence of flaxseed and lignans on colon carcinogenesis and beta-glucuronidase activity.

Flaxseed, the richest source of mammalian lignan precursors, such as secoisolariciresinol diglycoside (SD), has been shown over the short term to decrease some early markers of colon cancer risk. This study determined whether over the long term flaxseed still exerts a colon cancer protective effect, whether its effect may, in part, be due to its high content of SD and whether any change in beta...

Hypocholesterolemic and antiatherosclerotic effect of Flax lignan

Hypercholesterolemia, low HDL-C and oxygen radicals have been implicated in the development of atherosclerosis. Lignan complex isolated from Flaxseed (Linum usitatissimum Linn., Linseed, Hindi ⎯ Alsi) contains secoisolariciresinol diglucoside (SDG), 3-hydroxy-3-methylglutaric acid (HMGA) and cinnamic acids. SDG and cinnamic acids are antioxidants and HMGA is a hypocholesterolemic agent. Antioxi...

Dietary flaxseed lignan extract lowers plasma cholesterol and glucose concentrations in hypercholesterolaemic subjects.

Lignans, derived from flaxseed, are phyto-oestrogens being increasingly studied for their health benefits. An 8-week, randomised, double-blind, placebo-controlled study was conducted in fifty-five hypercholesterolaemic subjects, using treatments of 0 (placebo), 300 or 600 mg/d of dietary secoisolariciresinol diglucoside (SDG) from flaxseed extract to determine the effect on plasma lipids and fa...

Health effects with consumption of the flax lignan secoisolariciresinol diglucoside.

Flaxseed is the richest source of the lignan secoisolariciresinol diglucoside (SDG). After ingestion, SDG is converted to secoisolariciresinol, which is further metabolised to the mammalian lignans enterodiol and enterolactone. A growing body of evidence suggests that SDG metabolites may provide health benefits due to their weak oestrogenic or anti-oestrogenic effects, antioxidant activity, abi...
